IIRC, the 22/45 has a polymer frame while the Mark series is all steel. I would go for the Mark III personally.

Re: ruger mkiii or 22/45
The MIII is a cadillac, the 22/45 is a Chevy, that is not to say the 22/45 is not a fantastic gun afterall the Corvette is a Chevy.
THe biggest difference is in the grip and angle of the grip , the MI, MII and MIII is patterened after a P1908 Luger, the 22/45 is patterned after a 1911 hence the 45 designation.
Try both and see which one you like best.
Some people do not like the grip angle on the MIII, so try before you buy.

I've got a 22/45 and love it. I didn't care for the high angle of the MkIII.

Re: ruger mkiii or 22/45
My wife has a 22/45 with removable panels and the grip angle is very similar to a 1911 which makes it comfortable to shoot for rimfire competitions. I have a MkII and while it feels more substantial and solid as compared to my wife's 22/45, I find that during competition, the higher grip angle causes my hand to get in the way of the "slide." When I'm shooting at the range, it's not an issue because I adjust my grip to prevent the web of my hand from getting in the way, but under competition stress I find myself going towards the same grip as I would use on my 1911 and I get slide bite.
So I guess, for me at least, it would depend on what you plan on using the gun for. Honestly, they're both great guns, I just think the 22/45 works better for competition.
Oh, and my wife's 22/45 - bull barrel - came with a scope rail.

To avoid the "sometimes you feel like a nut, sometimes you don't" dilema,do what i did,get one of each, you just can't go wrong.
Government 99 and 44/100 % pure bullshit.

I have the Bull barrel target model blued; Either would work I do lean toward the BB target over the narrow barrel models.
The 22/45 is suppose to give the feeling of holding/shooting a 1911, if you can get into that. Both recievers are nearly the same and should give a life time of use as is Rugers call to fame.

Re: ruger mkiii or 22/45
22/45 is a polymer lower and a 1911 grip angle. I went with this one for lighter weight and because i prefer that grip angle. the mark III's grip angle just isn't preferable to me. this and the metal lower are the main differences. both bull barrels of 5.5 inches come with scope/optic rails and are already drilled and tapped. the mark III series, (to include the 22/45 mk III) however has a mag disconnect safety that will have to be swapped with a mark II bushing in order to have the magazines drop free. you can also do the slingshot mod and grip pannel mod (22/45) since. i purchased mine before ruger made the model with interchangeable grip pannels. so just figure out which grip angle you prefer. if you like 1911's the 22/45 at $240 is the way to go.
